Why are my PCR tubes crushed/deformed after my PCR run?

The heated thermal cycler lid is designed to make sure that optimized pressure is applied on the microplate for an efficient PCR reaction. When using PCR tubes, excess pressure on the tubes, from the heated lid can cause deformation of the tubes. To avoid this, we recommend using the Tray/Retainer set, as shown below:

To add an infinite hold, set the time for the hold to ‘99.59'. The screen should then default to the infinity symbol for your hold.

Is the GeneAmp PCR System 9700 capable of running a gradient?

No, the GeneAmp PCR System 9700 does not have a gradient feature. Our newer thermal cyclers including the ProFlex PCR System and Veriti Thermal Cycler have better-than-gradient VeriFlex Blocks.

What is the Auto Delta (AutoX) feature on the GeneAmp PCR System 9700?

The AutoDelta feature allows you to incrementally increase or decrease the temperature or hold time with each cycle for a given step within a cycling stage. One common application for this is touchdown PCR.

How do I use the adaptor and compression pad for Applied Biosystems OpenArray off-line cycling on the Applied Biosystems GeneAmp PCR System 9700?

- Position the adaptor.
- Insert the two adaptor pins into the circular slots on the module.
- Be sure the adaptor sits flat.
- Position the Applied Biosystems TaqMan OpenArray plates.
- Orient each plate with the barcode facing up and along the edge of the block, away from the center of the adaptor.
- Fill up the right sample block first when placing the adaptor on the module (right sample block is the primary control block).
- Fill up the center slots before any edge slots.
- (Recommended) Insert unused TaqMan OpenArray plates into any empty slots.
- Place the compression pad over the TaqMan OpenArray plates (only needed for Applied Biosystems NT-OpenArray plates, not for Applied Biosystems QuantStudio 12K), with the Teflon coating (smooth side) facing upwards and away from the plates.
- Slide the heated cover over the compression pad, then pull down the handle.

I have an Applied Biosystems GeneAmp PCR System 9700. What additional accessories do I need to perform off-line cycling for Applied Biosystems QuantStudio OpenArray plates?

You need an Applied Biosystems OpenArray Plate Adaptor (Part Number 4426824). You do not need a compression pad.

How can I get additional throughput for the Applied Biosystems QuantStudio 12K Flex OpenArray System?

Additional Applied Biosystems OpenArray plates can be run off-line using a flat-block thermal cycler. The two thermal cyclers we recommend are the Applied Biosystems GeneAmp PCR System 9700 and the Applied Biosystems ProFlex PCR System.
