Description

The 5500 SOLiD™ Fragment Library Core Kit is used to construct fragment libraries for sequencing on the 5500 Series Genetic Analysis Systems.

The 5500 SOLiD™ Fragment Library Core Kit contains the necessary reagents (enzymes, buffers, and PCR Amplification mix) for preparing 12 fragment libraries. Use in conjunction with either the 5500 SOLiD™ Fragment Library Standard Adaptor kit (PN 4464411) or the appropriate 5500 SOLiD™ Fragment Library Barcode Adaptor kit, which should be ordered separately.

The Core Kit consists of the 5500 SOLiD™ Fragment Library Enzyme Module (PN 4464413) and the 5500 SOLiD™ Fragment Library Amplification Module (PN 4464414).

Fragment libraries consist of short lengths of DNA specifically selected for optimal performance with a next generation sequencing platform for applications such as targeted or whole genome resequencing. Briefly, the 5500 SOLiD™ fragment library preparation protocol takes DNA through shearing, end-repair, A-tailing, and ligation with only one intervening bead-based purification that doubles as a size selection step. In many cases, subsequent library amplification is not needed, allowing high complexity libraries to be brought directly into the clonal amplification step.

Fragment libraries can be prepared in about four hours (or about five hours with amplification).
