My tubes are melting. What is wrong?

This can happen if the wrong plastics are used, or if the tubes are not being used with the correct tray/retainer set. Check the manual for directions on appropriate loading of the sample. Also, you can check for compatible plastics from here (https://www.thermofisher.com/us/en/home/life-science/pcr/pcr-plastics.html).

How do I program an infinite hold in the ProFlex PCR System?

To add an infinite hold, select the infinity symbol for program.

How do the VeriFlex Blocks work on the ProFlex PCR System?

In the ProFlex PCR System, the 96-well and 3 x 32-well blocks feature innovative VeriFlex Blocks for enhanced PCR functionality. Six separate Peltier blocks (2 on each 32-well block for 3 x 32-well) provide maximal versatility and flexibility and offer two key benefits:

1.PCR optimization: each block can be set with a specific temperature, which is ideal for precise control over PCR optimization.
2.Run more experiments: the six Peltier blocks can also be used to run up to six different annealing temperatures in the same run.

What is the Auto Delta feature on the ProFlex PCR System?

The AutoDelta feature allows you to incrementally increase or decrease the temperature or hold time with each cycle for a given step within a cycling stage. One common application for this is touchdown PCR.

What is the ramp rate of the ProFlex PCR System?

The 96-well and 3 x 32-well system have a max block ramp rate of 6.0 degrees C/sec and the dual 96-well and dual flat blocks have a max block ramp rate of 2.0 degrees C/sec.

What types of plastics can I use with the ProFlex PCR System?

The plates and tubes will be specific to the block type. Please refer to our PCR Tubes, Plates & Accessories Plastics Selection Guide (https://www.thermofisher.com/us/en/home/life-science/pcr/pcr-plastics.html) for a list of compatible plastics, based on your block type.

What types of blocks are available on the ProFlex PCR System?

The ProFlex PCR System has 4 different blocks that can be changed with the flip of a switch, including the first-of-its-kind 3 x 32-well independent block. This block allows up to 3 experiments to be run simultaneously, completely independently of each other and without interference. A dual 96-well block is available for your high throughput sequencing needs and a dual flat block is available to support OpenArray plate technology for genotyping analysis on the QuantStudio 12K Flex Real-Time PCR System.

What is the optimal ramp rate for my thermal cycler?

There is no universal ramp rate for a thermal cycler because both the block and sample ramp rates are different for each thermal cycler. The sample ramp rate changes with the volume of the sample since more time and energy are required to heat higher sample volumes. The ramp rate may also need to be changed for a particular PCR assay or primer pair if you are trying to use the same PCR assay on two different thermal cyclers, for example.

As a reference, the block ramp rate commonly used with DreamTaq Polymerase on the MiniAmp thermal cycler is 3.0 C/sec. In a 50 µL PCR reaction, this translates to a sample ramp rate of ~1 C/sec.

The ProFlex PCR System can be connected wirelessly with an optional High-Power USB Wi-Fi Module, Cat. No. A26774. Please note that you will need to update to the latest version of firmware in order to use this Wi-Fi module.

How can I get additional throughput for the Applied Biosystems QuantStudio 12K Flex OpenArray System?

Additional Applied Biosystems OpenArray plates can be run off-line using a flat-block thermal cycler. The two thermal cyclers we recommend are the Applied Biosystems GeneAmp PCR System 9700 and the Applied Biosystems ProFlex PCR System.
