With the PrioCHECK™ M. avium Antibody ELISA appropriate information about the MAA status in pig herds can be obtained and, based on the results, corrective measures to improve the MAA status in the pig herd can be implemented. In a risk-based surveillance scheme using the PrioCHECK™ M. avium Antibody ELISA, only a fraction of the pigs needs to be tested. This saves time and labor as no incision of the lymph nodes of each individual animal at slaughter needs to be made. The test delivers more accurate results of infections at the herd level, allowing efficient implementation of appropriate control measures.
Test procedure
The PrioCHECK™ M. avium Antibody ELISA is a diagnostic kit based on ELISA technology. It follows a four step protocol consisting of sample preparation, sample incubation, conjugate incubation and detection. Serum or meat juice samples are incubated in an ELISA plate coated with the polar lipid fraction antigen of M. avium subsp. hominissuis. Anti-M. avium antibodies present in the sample bind to the antigen. Plates are then washed and an anti-swine antibody labeled with horse radish peroxidase (HRPO) is used to detect antibodies bound to the M. avium antigen. Color development using tetramethyl-benzidine (TMB) substrate measured optically at a wavelength of 450 nm shows the presence of antibodies directed against Mycobacterium avium.
