How can I test my tissue for the presence of endogenous biotin?

Deparaffinize and rehydrate the paraffin-embedded sections and then incubate with streptavidin-HRP followed by DAB or AEC. Perform this test only after removal of endogenous peroxidase activity to rule out background staining caused by endogenous peroxidase. High endogenous biotin in mast cells, kidney, liver, pancreas, intestine, spleen, and other tissues can cause high background staining with a streptavidin-biotin-based detection system. In such cases, our SuperPicTure Polymer Detection Kit (Cat. No. 878963) is recommended because it does not include streptavidin or biotin. If needed, these staining kits (Cat. Nos. TL-015-HA, TL-015-HAJ, TL-015-HD, and TL-015-HDJ) can also be used since they do not contain streptavidin or biotin.
