What will happen if the MagMAX HMW DNA Booster is not added during the workflow?

The isolated DNA recovered would likely not achieve greater than 100 kb in fragment size and the workflow reproducibility of the HMW kit would be compromised. If the customer wishes to leave the Booster Solution out of the workflow, we recommend following the guidance of Applied Biosystems(TM) MagMAX(TM) DNA Multi-Sample Ultra 2.0 kit to achieve genomic DNA without expanding into the HMW (>100 bp) or Ultra-HMW DNA (around 300 bp) recovery range.

Why is it recommended to use a mixture of wide bore and standard P1000 pipette tips for the MagMAX HMW DNA workflow?

Fragile samples can get damaged when they are forced through the narrow orifice of standard pipette tips. Using a combination of standard and wide bore pipette tips helps to increase the yield of HMW DNA, keeping the fragment sizes intact. Reference the MagMAX HMW DNA User Guide (MAN1001478).

Are low-binding tubes essential for manual workflows?

Yes, low-bind tubes are recommended to minimize loss due to absorption to the tube walls. Please refer to MagMAX HMW DNA User Guide (MAN1001478) for guidance on when to transfer to new tubes to reduce risk to sample contamination.

Why does the MagMAX HMW DNA User Guide specify a step-by-step reagent addition when preparing the sample plate for blood and cell samples, but recommends a master mix (pre-prepared mixture of reagents) for tissues?

The enhancer solution improves proteinase K activity at optimal concentrations; however, excessive enhancer solution can reduce proteinase K effectiveness. For blood and cell samples, adding the blood sample and cell lysate in the correct order dilutes the enhancer to achieve optimal proteinase K activity during the digestion step. For tissue samples, the tissue must be digested with proteinase K before using the tissue lysate for extraction. Preparing a master mix with Cell/Tissue extraction buffer ensures the enhancer solution is diluted to the concentration needed for optimal proteinase K activity in these samples.

Is it necessary to use all wells within the KingFisher 96 deep-well plate?

No, it is not necessary to fill all wells of a KingFisher 96 deep-well plate. If you are using less than the full-plate format, simply don't add any reagents to the unused wells for the isolation workflow.
