The Gibco Hepatic Spheroid Kit is the first off-the-shelf kit for generation of hepatic spheroids in your own lab.
•
Maintain cells in culture longer—up to 21 days, >4X longer than cells last in monolayer culture
•
Design your own schedule—cells are ready for plating when you are
•
Interrogate pathways of interest—applicable for toxicity, metabolism, or disease modeling
What you get:
• Spheroid-qualified hepatocytes
• Hepatocyte Thaw Medium
• Plating supplement (Cocktail A + Dexamethasone)
• Maintenance supplement (Cocktail B + Dexamethasone)
• Fetal bovine serum
• Williams' E Medium, no phenol red
• Nunclon Sphera 96-well U-bottom plates
Please note that components are shipped separately due to differences in storage temperatures.
Simplified protocolCryopreserved cells can be thawed and plated at your convenience, in as little sa 60 minutes. Try it on your own, or
request a demo with an experienced scientist.
Stringent quality controlOur high standards help ensure only the best spheroid-forming lots become available, maximizing your chance of success. In addition, each lot is tested for:
• Albumin production in monolayer and spheroid culture
• Phase I enzyme induction
• Bile duct formation (CFDA staining)
• Proper morphology
• Viability >75%
• Attachment efficiency
• Monolayer formation and integrity
Important! For best results, do not attempt hepatic spheroid formation in low attachment plates from other sources. Hepatocyte lots are qualified in Nunclon Sphera plates only and use of other plates can lead to variability in spheroid formation.
Flexible readoutsAnalyze single spheroids, or pool several for detection in assays such as gene expression, immunostaining, viability, or CYP activity.
