The Annexin V-FITC Apoptosis Detection Kit has a different protocol because it has been validated in-house with that protocol and as a kit. The protocol, for the Annexin V Recombinant Protein FITC, is a generic staining protocol that generally requires optimization per application. Essentially the ideal titration would need to be determined empirically and specifically for your instruments.

The Annexin V-FITC Apoptosis Detection Kit protocol is more scrutinized and validated to work as an indicator of apoptosis, whereas the Annexin V Recombinant Protein FITC by itself is validated to bind to phosphatidylserine on cells. The standalone product allows users to co-stain with non-PI counterstains, or use it in other applications aside from traditional apoptosis.

For this product, a test is defined as 1x10^6 cells (1million) in 100 µl of staining volume, so specifically 5 µl of the Annexin V FITC dye is enough to stain 1x10^6 cells in 100 µl.
