Es bildet problemlos ein gemischtes Disulfid mit Thiolen, das die Chromophore 5-Merapto-2-Nitrobenzoesäure (Absorptionsmaximum 410 nm) freisetzt. Nur Protein-Thiole, die für dieses wasserlösliche Reagenz zugänglich sind, werden modifiziert.
