Specificity: no significant cross-reaction with mouse IL-2, IL-3, IL-4, IL-5, IL-6, GM-CSF, IFNγ or human TNFα. This ELISA has some cross-reactivity with rabbit and cynomologous monkey TNFα in culture supernatants.

This mouse TNF alpha ELISA kit uses pre-titered, matched pairs of coating and detection antibodies to achieve sensitive and accurate measurement of tumor necrosis factor alpha (TNF-alpha, TNFα) in samples at 50 to 100 µL per well. Also included are calibrated recombinant protein standard, streptavidin-HRP, TMB substrate solution and stop solution. The complete kit contains a precoated 96-well strip plate and lot-optimized reagents. All components undergo quality control testing to minimize lot-to-lot variability.

TNFα, also called cachectin, macrophage cytotoxin, necrosin, etc., is produced primarily by macrophages in response to bacteria, viruses, mycoplasma, immune complexes, other cytokines, tumor cells, complement and phorbol esters among others. It is a 157 amino acid long polypeptide with an apparent MW of 17 kDa under denaturing conditions but exists as a 45-55 kDa trimer naturally.

In vivo, TNFα has anti-tumor activities against some solid tumors, induces tumorigenesis, metastisis; acts in immunomodulation; induces septic shock, fever; plays a role in multiple sclerosis; and is involved in autoimmunity and infectious disease. TNFα alone and in combination with other cytokines and chemical agents has been used as an anti-tumor agent. Patients with melanoma and other solid tumors, administered high doses of TNFα, have realized a cure rate of up to 90% albeit with some major and minor side effects.
