The His-Patch ThioFusion™ Expression System improves upon the original ThioFusion™ System by eliminating the need for specialized E. coli strains and media, allowing easier subcloning of the gene of interest, and simplifying purification of fusion proteins. The vector used for expression, pThioHis, contains a modified thioredoxin gene that allows purification of fusion proteins on metal-chelating resins (i.e., ProBond™ resin). When the modified thioredoxin protein, His-Patch (HP) thioredoxin, is expressed, a histidine patch is formed. This histidine patch has a high affinity for divalent cations (i.e., nickel) allowing simple, rapid purification on metal-chelating resins such as ProBond™. pThioHis also encodes an enterokinase cleavage site for efficient cleavage of N-terminal fusions with EKMax™ enterokinase.

The system contains 20 µg each of pThioHis A, B, & C vectors, ProBond™ resin, columns, and buffers for six purifications, TOP10 E. coli stab, Trx forward and reverse sequencing primers. Store lyophilized and resuspended vector at -20°C. Store ProBond™ resin at +4°C. Store all other components at room temperature. All reagents are guaranteed stable for 6 months when properly stored.
