This recombinant fusion protein consists of full-length glycogen synthase kinase 3 alpha (GSK3A) and a glutathione S-transferase (GST) tag. It is validated in the LanthaScreen® Eu Kinase Binding Assay. No special measures were taken to activate this kinase. • Compatible and validated with the LanthaScreen® Eu Kinase Binding Assay • Expressed in insect cells • GST tagged for use with Eu-labeled GST antibody
Easily Screen for Kinase Binding The LanthaScreen® Eu Kinase Binding Assay is a TR-FRET assay that lets you measure inhibitor binding to non-activated kinases, perform simple off-rate measurements and makes simple the study of type III (allosteric) inhibitors.
The LanthatScreen® Eu Kinase Binding Assay is easy to optimize and run. The assay uses a simple mix-and-read format with a single one hour incubation and no development steps. Simultaneous binding of an Alexa Fluor® conjugate or tracer” to a tagged kinase is detected by addition of a europium-labeled anti-tag antibody, in this case anti-GST. Simultaneous binding of the tracer and antibody to the kinase results in a high degree of fluorescence resonance energy transfer (FRET), while displacement of the tracer with a kinase inhibitor results in a loss of FRET (Fig 1).
GSK3A for the LanthaScreen Kinase Binding Assay Glycogen synthase kinase 3 alpha (GSK3A) is a multifunctional protein serine/threonine kinase implicated in the control of several regulatory proteins and transcription factors. (GSK3A Alias: DKFZp686D0638)
This insect cell-expressed recombinant fusion protein of full-length GSK3A and GST tag has been validated in the LanthaScreen® Eu Kinase Binding Assay with these reagents: • Tracer 236 (Cat No. PV5592) • Eu-labeled GST Antibody (Cat No. PV5594)
