Detect the deoxyribonuclease (DNase) activity in microorganisms with Thermo Scientific™ Remel™ DNase Test Agar w/Methyl Green.
In 1957, Jeffries et al. developed a medium to demonstrate DNase activity of microorganism that possess this extracellular enzyme1. This medium is used to identify a variety of bacteria including staphylococci, enteric gram-negative bacilli and pseudomonads2-5. On DNase Test Agar w/Methyl Green, the colony is surrounded by a clear zone in an otherwise green medium6.
DNase Test Agar w/Methyl Green is a solid medium used to detect the deoxyribonuclease (DNase) activity in microorganisms. For the methyl green modification, the dye methyl green is able to bind to polymerized DNA to form a stable green complex at pH 7.3. The action of DNase depolymerizes the DNA structure releasing methyl green; when not combined at this pH methyl green becomes colorless resulting in colorless halos around DNase-positive colonies.
DNase Test Agar w/Methyl Green contain casein and soy peptone that supply nitrogen, amino acids and peptides necessary for bacterial growth. Sodium chloride provide essential electrolyte which maintain osmotic equilibrium.
