Middlebrook 7H10 Agar is a solid medium that allows faster and luxurious growth of Mycobacteria1 and protects the organisms against toxic agents2. The Middlebrook 7H11 Agar, developed by Cohn et al., contains casein hydrolysate that stimulates growth of drug-resistant strains of M. tuberculosis. Mitchison et al. modified this formulation by adding antibiotics as selective agents with the 7H11 base3 to selectively isolate Mycobacteria.

Middlebrook 7H11 Agar/Mitchison 7H11 Selective Biplates contain inorganic salts essential for growth of mycobacteria. Casein hydrolysate stimulates growth of drug resistant strains of Mycobacterium tuberculosis4. Glycerol is the source of carbon and energy. Sodium citrate is converted to citric acid which holds inorganic cations in solution.
OADC Enrichment is a supplement which contains following additives: sodium chloride to maintain osmotic equilibrium, dextrose that serves as an energy source, catalase to destroy toxic peroxides that may be present in the medium; oleic acid which is required in the metabolism of mycobacteria; and albumin to protect tubercle bacilli against toxic agents. It also contains selective agents malachite-green dye, polymyxin B, carbenicillin, amphotericin B and trimethoprim. Polymyxin B and carbenicillin are selective agents active against most Enterobacteriaceae. Amphotericin B and trimethoprim are active against yeast and Proteus spp., respectively.
