Selectively isolate and differentiate coliform organisms and cultivate a wide variety of microorganisms to visualize hemolytic reactions with Thermo Scientific™ Remel™ Tergitol 7 Agar with TTC/Blood Agar Biplates. Tergitol 7 Agar was developed by Chapman for selective isolation and increased recovery of coliform organisms2,3 and further modified by addition of triphenyltetrazolium chloride (TTC) to facilitate early recognition of E. coli and E. aerogenes. Blood agar consists of tryptic soy agar with blood to facilitate growth of fastidious bacteria and observe hemolytic reactions.
Selectively isolate coliform organisms, cultivate a wide variety of microorganisms and visualize hemolytic reactions using the solid medium Tergitol 7 Agar with TTC/Blood Agar Biplates. The convenient biplate format contains Tergitol 7 Agar w/TTC in one section and Blood Agar in the other section.
Tergitol 7 Agar w/TTC facilitates early recognition of Escherichia coli and Enterobacter aerogenes within 6-10 hours of incubation. TTC incorporated into Tergitol 7 Agar is a rapid indicator of bacterial growth. Some coliforms reduce TTC to insoluble formazan resulting in red colonies with blue zones that appear as purple. E. coli and E. aerogenes do not reduce TTC and form yellow colonies with yellow zones.
Blood Agar (TSA w/5% Sheep Blood) is suitable for the cultivation of fastidious aerobic and anaerobic bacteria, the latter of which can be grown in either deep cultures or by incubation under anaerobic conditions. It facilitates rapid production of large colonies, clearly defined zones of hemolysis and good colonial differentiation. Blood Agar is suitable for the CAMP test.
In the Blood Agar medium, Group B streptococci produce a protein-like compound called CAMP factor that acts synergistically with beta lysin to produce an arrowhead shaped area of complete lysis4.
Tergitol 7 Agar w/TTC contains yeast extract, casein and meat peptone that provide nitrogen, amino acids and peptides necessary for bacterial growth. Lactose serves as the source of fermentable carbohydrate. Tergitol-7 acts as a selective agent that inhibits gram positive organisms and minimises the swarming of Proteus, allowing superior recovery of coliforms. Brom thymol blue is the pH indicator.
Tri-phenyltetrazolium chloride (TTC) allows earlier recognition and identification of Escherichia coli and Enterobacter aerogenes.
Blood Agar contains casein and soy peptones which provide nitrogen, amino acids and peptides necessary for bacterial growth. Sheep blood provides essential growth factors which enrich the medium and allows hemolytic reactions to be demonstrated.
