Determine yeast fermentation reactions using Thermo Scientific™ Remel™ Yeast Fermentation Broth with Brom Thymol Blue (BTB) and Carbohydrates. Wickerham developed a chemically defined broth medium to determine yeast carbohydrate assimilation reactions which occur in the presence of oxygen1. Fermentation reactions, on the other hand, occur in the absence of oxygen and are evidenced by gas formation. Aheam and Haley used various carbohydrates in a modification of Wickerham’s formula to determine the fermentation reactions of yeast isolates2,3.

Yeast Fermentation Broth with BTB and Carbohydrates is a liquid media recommended for ascertaining yeast fermentation reactions. Fresh and processed products including heat-processed foods are subject to spoilage. Certain foodborne yeasts may also elicit allergic reactions or may cause infections. Carbohydrate fermentation tests provide a useful means for yeast identification when other tests fail to identify an isolate.
This medium contains peptone that provides nitrogenous compounds and amino acids essential for the growth of yeasts. Yeast extract supplies B-complex vitamins. Brom thymol blue is used as a pH indicator. The anaerobic utilization of a carbohydrate results in the formation of ethanol and carbon dioxide. The following carbohydrate combinations are available: Cellobiose, dextrose, galactose, lactose, maltose, raffinose, sucrose, trehalose, xylose
A positive fermentation reaction is indicated by the presence of gas bubbles in the inverted Durham tube. The development of a yellow color in the fermentation test (acid pH) indicates the carbohydrate has been assimilated.
