Cultivate and isolate Mycobacterium species with Thermo Scientific™ Remel™ Middlebrook 7H10 Agar. Based on the formulations developed initially by Dubos and Middlebrook1, and modified by Middlebrook and Cohn2, the Middlebrook 7H10 agar promotes the faster growth of Mycobacterium species and protects tubercle bacilli against a variety of toxic agents1.
Middlebrook 7H10 Agar is a solid medium recommended for use in qualitative procedures for the cultivation and isolation of mycobacteria. Mycobacteria are immobile, slow-growing, rod-shaped, gram-positive bacteria with high genomic content. The genus includes pathogens known to cause severe diseases in mammals, including tuberculosis and leprosy.
This medium contains inorganic salts which are essential to the growth of mycobacteria. Glycerol is a source of carbon and energy. Sodium citrate is converted to citric acid, which holds inorganic cations in solution. The malachite-green dye is a selective agent, which partially inhibits bacteria other than mycobacteria. Oleic Albumin Dextrose Catalase (OADC) Enrichment is a supplement which contains the following additives: sodium chloride to maintain osmotic equilibrium; dextrose, an energy source; catalase to destroy toxic peroxidases that may be present in the medium; oleic acid, which is required in the metabolism of mycobacterium; albumin to protect the tubercle bacilli against toxic agents.
