Rapidly identify Group A Streptococci and Enterococci using Thermo Scientific™ Remel™ PYR Reagent.In 1981, Godsey, et al differentiated group A streptococci and enterococci based on their ability to cleave L-pyrrolidonyl-β-naphthylamide (PYR)1. Facklam et al. incorporated PYR substrate into agar and tested for enzyme activity after overnight incubation2. Bosely et al. incorporated PYR substrate into broth and tested for enzyme hydrolysis after 4 hours incubation3. Ellner et al. described a colorimetric method for the PYR test using filter paper strips containing PYR substrate4.

Use PYR Reagent for rapid identification of Group A Streptococci and Enterococci.Rapid Test - Color change observed within 1-2 minutes after addition of reagentReady-to-use - Liquid reagentEasy to interpret -Red color developmentL-pyrrolidonyl-β-naphthylamide (PYR) serves as the substrate for the detection of pyrrolidonyl peptidase. The peptidase enzyme hydrolyzes the substrate to beta naphthylamine which produces a red color on addition of N,N-dimethyl-aminocinnamaldehyde (PYR reagent). The ease of performance and interpretation of the PYR test suggests that it is more specific than the routine bacitracin test for group A Streptococci and is at least as specific as the routine 6.5% NaCl test for Enterococci5.This reagent contains 0.15 g/1000mL of N,N-dimethyl-aminocinnamaldehyde (PYR reagent).Not all products are available for sale in all territories.
