Thermo Scientific™ Remel™ Absolute Methanol is used with the modified acid-fast stain procedure for detection of Cryptosporidium in clinical specimens. Ma and Soave recommended a modified acid-fast staining procedure because of its simplicity and specificity for detecting Cryptosporidium1. Garcia recommended a more recent modification of the Kinyoun carbolfuchsin stain that is used to detect Cryptosporidium oocysts in clinical specimens 2,3. Absolute Methanol is a fixative used to preserve the morphology of the cells and aids in detection of Cryptosporidium oocysts.
Cryptosporidium is an intestinal protozoan parasite well known for causing diarrhoea in animals and has more recently been recognized for causing human disease. Oocysts are spherical 4-6μm in diameter, and stain pink to red to purple with Kinyoun carbolfuchsin stain. Most will contain several dark staining granules.
