Gibco CTS Detachable Dynabeads CD4 are intended for ex vivo isolation of CD4+ T cell subsets. Combined with CTS Detachable Dynabeads Release Buffer (Cat. No. A5588301, A5588302, or A5588303), it is a first-of-its-kind cGMP cell selection technology with an active release mechanism for process development, clinical trial, and commercial manufacturing use. This product is also commonly used with CTS Detachable Dynabeads CD8 (Cat. No. A56995).

Features of CTS Detachable Dynabeads CD4 include:

  • Empower users with process flexibility by allowing active release of beads from target cells at any time
  • Optimize target cell purity and yield thanks to the active release mechanism that gives users more control
  • Provide an automation-friendly solution by working directly with the CTS DynaCellect Magnetic Separation System
  • Enable consistent performance in light of biological variability within the starting material
  • Suitable for both autologous and allogeneic cell therapy manufacturing processes

These Detachable Dynabeads are intended for use with CTS Detachable Dynabeads Release Buffer, which has an active ingredient, biotin, that competitively displaces a biotin derivative on the surface of the beads. This proprietary mechanism allows users to achieve active release of the beads from target cells at any time within their process.

Based on a recommended 4-to-1 bead-to-target-cell ratio (not total nucleated cell number), one 15 mL vial can theoretically isolate up to 1.5 x 109 CD4+ T cells.

CTS Detachable Dynabeads also include a Drug Master File (DMF) to support cross-referencing in regulatory submissions prior to clinical trial and commercial manufacturing use.

For Research Use or Manufacturing of Cell, Gene, or Tissue-Based Products. CAUTION: Not intended for direct administration into humans or animals.

Frequently asked questions (FAQs)

What is the definition of superparamagnetic, and what does this mean for my cell isolation application with Dynabeads magnetic beads?

Superparamagnetic means that the Dynabeads magnetic beads exhibit magnetic properties when placed within a magnetic field, but have no residual magnetism when removed from the magnetic field.

This means that your targeted cells, proteins, or nucleic acids are only subjected to magnetic forces during the time the beads are on the magnet. The beads do not aggregate, but remain evenly dispersed in suspension.

What are the shipping and storage and conditions for CTS Detachable Dynabeads CD4 (Cat. No. A56994)?

CTS Detachable DynabeadsCD3/CD28 (Cat. No. A56996) is shipped at 2-8 degrees C. We recommend storing it at 2-8 degrees C, protected from light.

What are the antibodies on the surface of CTS Detachable Dynabeads CD4 and CTS Detachable Dynabeads CD8?

CTS Detachable Dynabeads CD4 and CTS Detachable Dynabeads CD8 are coated with camelid-derived, single-domain (VHH) antibody fragments that identify and bind CD4 or CD8 markers, respectively. These antibodies are produced in an Animal Origin Free (AOF) yeast-based system virtually eliminating any risk of viral contamination.

When using CTS Detachable Dynabeads CD4 or CTS Detachable Dynabeads CD8, what is the recommended ratio of Dynabeads to target T cells?

When using CTS Detachable Dynabeads CD4 or CTS Detachable Dynabeads CD8, we recommend a 4:1 ratio of Dynabeads to target T cells (CD4+ or CD8+ T cells, respectively). Please do not calculate it based on the number of total nucleated cells in your starting material.
