Invitrogen SuperScript IV Template-Switching RT Master Mix is designed for high template switching efficiency in cDNA synthesis reactions. It contains SuperScript IV Reverse Transcriptase, RNase Inhibitor, and all other reverse transcription reaction components in a convenient premixed format. Only RNA template, RT primer, and template switching oligo (TSO) of choice need to be added.

Procedure overview
To maximize template switching efficiency, SuperScript IV Template-Switching RT Master Mix contains proprietary template switching enhancer and leverages the terminal deoxynucleotidyl transferase (TdT) activity of SuperScript IV Reverse Transcriptase. When the reverse transcriptase reaches the 5’ end of the RNA template, TdT activity adds 1–3 extra nucleotides to the cDNA end, enabling binding of the template switching oligo and incorporation of known sequence at the 3’ end of the resulting cDNA. This cDNA can be used for amplification by PCR, gene expression analysis by real-time PCR, or as a template for 5’RACE, RNA-seq, or second strand cDNA synthesis.This master mix is compatible with a wide input range of total RNA (from 2 pg to 4 µg) and is supplied with lysis buffer to enable direct cDNA synthesis from mammalian cells (1–1,000 cells per sample).

Invitrogen Template Switching Oligo (Cat. No. A65427) is offered, in conjunction with Capturing Oligo (dT) (Cat. No. A65429) and Preamplification Primers (Cat. No. A65428), complementary to the TSO. Alternatively, custom self-designed template switching oligo can be used.

Can SuperScript IV Template Switching RT Master Mix be used for other applications that do not involve template switching, e.g. RT-qPCR?

Yes, the master mix can be used for the first strand cDNA synthesis without templates switching oligo, but the results may be not as good as with more specialized product. For RT-qPCR, we recommend SuperScript IV VILO Master Mix.

Lysis buffer supplied with SuperScript IV Template Switching RT Master Mix has a dual function. It enables direct cDNA synthesis workflow without RNA purification, and it includes additional amount of proprietary enhancer which improves templates switching efficiency with low input samples of purified RNA (up to 10 ng).
