The Image-iT Low Density Lipoprotein Uptake Kit - Bodipy FL includes Bodipy FL dye-labeled LDL, unlabeled LDL, heparin, BSA, and assay buffer. The kit was designed to specifically detect binding and uptake of LDL through the LDL receptor internalization pathway, allowing maximum control and flexibility in the experimental design. It can be used with fluorescence microscopy, flow cytometry, cell sorting, high-throughput analysis, and high content analysis.

• Optimized LDL uptake kit with two built-in controls
• Bodipy FL is detectable with GFP/FITC filter set (absorption peak: ∼515 nm, emission peak: ∼520 nm)
• Bodipy FL is fixable and mutiplexable with orange, red, deep red, violet, and blue emission dyes, such as RFP, mCherry, DAPI, Hoechst, HCS LipidTOX, and LysoTracker Deep Red

Two controls are included in the kit: unlabeled LDL as an optional pretreatment to occlude cell surface receptors in advance of probing with the labeled construct, and heparin which will chelate the LDL in the solution phase outside of the cells, preventing binding and uptake. When cells are labeled in the absence of these pretreatment controls, a fluorescent signal indicates specific LDL receptor-mediated endocytosis.

Frequently asked questions (FAQs)

Can the LDL in BODIPY FL dye-labeled LDL be oxidized?

We do not recommend doing this as the oxidation process (such as oxidation with copper sulphate) may result in the degradation of the BODIPY FL dye.

Can I use methanol or acetone for fixation of cells stained with BODIPY FL LDL?

If the BODIPY FL LDL is applied to live cells first and then fixed, the use of methanol or acetone fixation is not recommended. Exposure to alcohols, acetone, and other organic solvents can dissociate the BODIPY FL dye from the LDL.

No. Exposure to detergents and surfactants can dissociate the BODIPY FL dye from the LDL.
