Yes, you should block the western blot membrane before incubating with Streptavidin-HRP Conjugate. Blocking is essential to reduce non-specific binding and background signal.
We recommend incubating the membrane in 10 mL blocking solution. Gently agitate using a rocker platform for 1 hour at room temperature and then proceed to washing the membrane (Step 5 of the protocol on page 5 of the user manual (https://assets.thermofisher.com/TFS-Assets/LSG/manuals/streptavidin_conjugates_man.pdf).
