RPMI 1640 Medium (Roswell Park Memorial Institute 1640 Medium) was originally developed to culture human leukemic cells in suspension and as a monolayer. RPMI 1640 medium has since been found suitable for a variety of mammalian cells, including HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes, and carcinomas. We offer a variety of RPMI 1640 modifications for a range of cell culture applications. Find the right formulation using the media selector tool.

Using RPMI 1640 Medium
RPMI 1640 medium is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 medium contains biotin, vitamin B12, and PABA, which are not found in Eagle's Minimal Essential Medium or Dulbecco's Modified Eagle Medium. In addition, the vitamins inositol and choline are present in very high concentrations. RPMI 1640 medium with GlutaMAX™ supplement minimizes toxic ammonia build-up and improves cell viability and growth in an easy-to-use format. RPMI 1640 medium contains no proteins, lipids, or growth factors. Therefore, RPMI 1640 medium requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). RPMI 1640 medium uses a sodium bicarbonate buffer system (2.0 g/L) and therefore requires a 5–10% CO2 environment to maintain physiological pH.

How light sensitive is RPMI 1640 media? Should I also be protecting it from LED light?

While we know that different wavelengths of light are worse than others for exposure, we would recommend as a best practice to protect the medium from all forms of light exposure including LEDs, as much as possible to ensure optimal performance, as several components within the medium are light sensitive, such as vitamins.

What is the density (g/L) for RPMI 1640 Medium?

We have specific gravity information for RPMI 1640 Medium: 1.006 kg/L. In this case, the specific gravity is the same as density as the solvent is water.

Will depletion, absence, or breakdown of essential growth-promoting components such as glutamine or growth factors reduce the growth rate of my culture?

Yes. If you suspect that this is the case, remove the medium and add fresh medium. Alternatively, you can supplement medium with growth-promoting components. It is also possible to substitute GlutaMax I or II for glutamine in the medium to prevent glutamine exhaustion.

Will the media formulations containing GlutaMAX supplement change with respect to L-glutamine content?

In all media containing GlutaMAX supplement dipeptides as a substitute for L-glutamine, concentration is equimolar with the L-glutamine in the original formulation.
