El tampón de lisis de IP es un reactivo de lisis de células completas de mamíferos basado en una formulación de tampón de ensayo de radioinmunoprecipitación (RIPA) modificado sin dodecilsulfato sódico (SDS). Este tampón de lisis de fuerza moderada solubiliza con eficacia las proteínas celulares, pero no libera ADN genómico ni altera los complejos proteicos como los tampones de RIPA comunes. El tampón de lisis de IP Pierce está especialmente formulado para ensayos de precipitación e inmunoprecipitación.

El tampón de lisis de IP Pierce es eficaz para la lisis de células de mamíferos cultivadas a partir de células sembradas y células sedimentadas de cultivos en suspensión. El tampón de lisis está optimizado para los ensayos de precipitación e inmunoprecipitación. También es compatible con otras aplicaciones, incluidos los ensayos de proteínas Thermo Scientific 660 nm y BCA Thermo Scientific Pierce, purificación de proteínas e inmunoensayos (por ejemplo, ensayos de inmunoadsorción enzimática [ELISA] e inmunotransferencia [Western blot]).

El tampón de lisis de IP Pierce se compone de 25 mM de Tris-HCl pH 7,4, 150 mM de NaCl, 1 mM de ácido etilendiaminotetracético (EDTA), 1 % de NP-40 y 5 % de glicerol. El tampón no contiene inhibidores de fosfatasas o proteasas. Sin embargo, es posible añadir inhibidores (como el cóctel inhibidor de fosfatasas o el cóctel inhibidor de proteasas Thermo Scientific Halt) antes del uso para evitar la proteólisis y mantener la fosforilación de las proteínas.

Pierce IP Lysis Buffer ships at ambient temperature and can be stored at room temperature. However, we recommend 4 degrees C for long term storage in order to prevent possible bacterial growth.

Which protein assays are compatible with the Thermo Scientific IP Lysis Buffer?

The Thermo Scientific IP Lysis Buffer is compatible with Thermo Scientific BCA Protein Assay (Cat. No. 23225) and Thermo Scientific 660 nm Protein Assay (Cat. No. 22660). It is not compatible with Thermo Scientific Coomassie Plus (Bradford) Assay (Cat. No. 23236) unless diluted 1:1 in TBS, PBS, or water.
