Características del kit de aislamiento de mitocondrias para células cultivadas:

El aislamiento de mitocondrias suele ser un proceso laborioso que requiere el procesamiento de una sola muestra con homogeneización Dounce. El kit de aislamiento de las mitocondrias utiliza un método no mecánico basado en reactivos que permite que varias (seis) muestras de células cultivadas se procesen simultáneamente en alrededor de 40 minutos. Los gránulos de células de mamíferos cultivados se lisan con delicadeza gracias a una formulación patentada que da como resultado el máximo rendimiento de las mitocondrias con el mínimo daño a la integridad.

En las instrucciones del producto se describen las pautas para optimizar la pureza frente a los parámetros de rendimiento. El kit también ofrece un procedimiento de homogenización Dounce, que permite recuperar el doble de mitocondrias que el método basado en reactivos. Ambos métodos utilizan centrifugación diferencial para separar las fracciones citosólicas y de la mitocondria con una microcentrífuga de sobremesa. Se completan en aproximadamente 40 minutos (tras la recogida de células). Can I further purify my crude mitochondrial fraction isolated using your Mitochondria Isolation Kit for Cultured Cells (Cat. No. 89874)?

To obtain a more purified fraction of heavy mitochondria, centrifuge the post-nuclear supernatant at 3000g rather than 12,000g. Western blot analysis of purified versus crude mitochondrial fractions prepared with this kit result in a > 50% reduction in cathepsin S (lysosomal protein) and PMP70 (peroxisomal protein) in the purified fraction.
