El ATP enjaulado con NPE es un análogo de nucleótido en el que el fosfato terminal se esterifica con un grupo de bloqueo, lo que hace que la molécula sea biológicamente inactiva. La fotólisis flash del grupo enjaulado con iluminación UV a 360 nm provoca una liberación rápida y altamente localizada del nucleótido libre en el sitio de iluminación.
