What is the advantage of FreeStyle Max Reagent over 293fectin Transfection Reagent?
Both FreeStyle Max Reagent and 293fectin Transfection Reagent provide similar levels of high transfection efficiency; however, FreeStyle Max Reagent has lower cytotoxicity and hence results in higher protein yields. Additionally, FreeStyle Max Reagent is animal-origin free.
Can FreeStyle 293-F cells grown in FreeStyle 293 expression medium be transfected with ExpiFectamine293 Reagent along with enhancers? Is so, will the transfection efficiency be higher than that with FreeStyle Max Reagent?
You should be able to use the ExpiFectamine293 Reagent for transfection of Freestyle 293-F cells grown in FreeStyle 293 Expression Medium; however, the enhancers will provide only a little boost in expression as they are designed to work with higher density cultures that FreeStyle 293 Expression Medium cannot support.
What is the recommended ratio of DNA and transfection reagent when transfecting Freestyle cells?
The optimal amount will vary, but we recommend starting with a 1:1 ratio of DNA and lipid. For example, 1.25 ug DNA and 1.25 ul of the reagent for one ml of suspension cells.
How does FreeStyle MAX reagent compare to Lipofectamine 2000 and 293fectin reagents?
Lipofectamine 2000 is designed for adherent cells. FreeStyle and 293fectin reagents are designed for suspended Hek293 and CHO cells. For transfecting 293 suspension cells, FreeStyle MAX reagent yields transfection efficiency comparable to Lipofectamine 2000 and 293fectin reagents. However, Lipofectamine 2000 and 293fectin reagents do not work well for CHO suspension cells.
