Soluble DynActivator™ CD3/CD28 Prototype

The Gibco Soluble DynActivator CD3/CD28 Prototype is a soluble protein-based reagent designed to activate and expand enriched human T cell populations. The reagent is recommended for use with CTS Detachable Dynabeads CD4 (Cat. No. A56994) and/ or CTS Detachable Dynabeads CD8 (A56995).

Features of the Soluble DynActivator CD3/CD28 Prototype include:

• Soluble protein-based reagent for T cell activation via CD3/CD28
• Ready to use reagent with straightforward volumetric dosing
• Removable by simple medium exchange (features a soluble protein back-bone)
• One vial activates and expands at least 1.5 x 10⁹ T cells
• Empowers users with process flexibility by allowing separate isolation and activation steps

The Soluble DynActivator CD3/CD28 Prototype can be sterile filtered, and excess (remaining) reagent can be removed from cell culture by simple medium replacement or by a washing step using the CTS Rotea Counterflow Centrifugation System, if needed. T cell expansion is achieved by culturing for up to 7–14 days. For longer cultivation, restimulation after 14 days may be necessary.

The recommended titers (product amounts per cell number and volume) have been found to efficiently stimulate CD3+ T cells; however, for special applications, it is recommended to further experimentally determine the optimal stimulation titer. Over-activation of T cells carries a risk of activation-induced cell death.

The Soluble DynActivator CD3/CD28 Prototype has been developed in combination with Thermo Fisher Scientific T cell culture media: CTS OpTmizer T Cell Expansion SFM (Cat. No. A1048501) or CTS OpTmizer One SFM (A5757201) supplemented with PeproGMP Human IL-2 Recombinant Protein (GMP200-02) or PeproGMP Human IL-7 Recombinant Protein (GMP200-07) and PeproGMP Human IL-15 Recombinant Protein (GMP200-15).
