CHO-S-SFM II, is a complete, serum-free, low-protein (less than 100 μml) medium optimized for the growth and maintenance of Chinese hamster ovary (CHO) cells in suspension culture for production of recombinant proteins. It is supplemented with hypoxanthine and thymidine (the non-supplemented equivalent is also available). This ready-to-use medium is suitable for the suspension culturing of CHO and other cells in batch, continuous, and perfusion culture systems. A CHO-S-SFM II medium Drug Master File (DMF) has been submitted to the FDA. Permission to cross-reference the Master File may be obtained by contacting Technical Services or your local Sales Representative.

This medium contains components of animal origin. If growth factors or hydrolysates, found in competitor media, are to be avoided, consider CD CHO, CD OptiCHO™ or CD FortiCHO™ media.

When to use CHO-S-SFM II Medium
• Your cells have been successfully cultured in CHO-S-SFM II medium in the past and your process is nearing a regulatory filing (DMF available)

When to Consider Another Gibco™ Medium

Consider CD FortiCHO™ medium when:
• The cell line you are using is a transfected CHO K1, GS CHO or CHO-S™ cell line
• Maximum batch culture cell densities and protein titers are needed
• You have time to adapt cells into a new medium in an effort to maximize titers
• Cell health at harvest is a priority for downstream processing

Consider CD OptiCHO™ medium when:
• You are using a transfected CHO cell line other than CHO K1, GS CHO or CHO-S™ cell lines
• Optimization of feeding strategies will be part of your base medium selection testing in an effort to maximize productivity
• Your CHO cell line is “finicky,” i.e. hard to grow or adapt

Consider CD CHO medium when:
• Your GS CHO cells are recommended to be grown in CD CHO medium
• Your cells have been successfully cultured in CD CHO medium in the past and your process is nearing a regulatory filing (DMF available)

Consider CD DG44 medium when:
• You use parental DG44 or DXB11 CHO cells (dhfr-)

Which serum-free medium should I select for my CHO cells?

Thermo Fisher Scientific offers multiple serum free media (SFM) and chemically defined (CD) media options for CHO cultures. For adherent cultures, CD CHO (10743-011) and CD OptiCHO medium (12681-011) are robust media; both are protein free and animal origin free (AOF). For adherent cultures, CD CHO-A (097-0182DJ) is a widely used protein free and animal origin free medium. There are other formulations available to suit other research needs; contact Technical Support for further information.
