어플리케이션:

second-strand cDNA 합성 중 mRNA 제거 (1). oligo(dT)기 존재하는 상태에서 mRNA에서 poly(A) 시퀀스 제거 (2). Oligodeoxyribonucleotide 유도 RNA 절단 (3).

기원:

플라스미드에서 E. coli RNase H gene을 발현하는 E. coli 에서 정제됨.

성능 및 품질 시험:

Ribonuclease, 비특이 endodeoxyribonuclease, 3´ 및 5´ exodeoxyribonuclease.

단위 정의:

한 단위는 37°C에서 20분 내에 3 H-labeled poly(A)• poly(dT)의 RNA 1nmol을 산성 수용성 물질로 가수분해합니다.

단위 반응 조건:

20 mM Tris-HCl (pH 7.5), 0.1 M KCl, 10 mM MgCl2 , 0.1 mM DTT, 5% (w/v) sucrose, 0.5 nmol 3 H-labeled poly(A)• poly(dT), enzyme: 50 μl 37°C에서 20분간.
