이들 system은 DNA polymerase I의 고농도 exo-Klenow fragment와 random primer를 통합해 효과적으로 게놈 표적을 라벨링하여 민감한 게놈 프로파일링 실험을 할 수 있습니다. Exo-Klenow는 5' 및 3' exonuclease 활성이 모두 제거된 DNA polymerase I holoenzyme의 큰 분절 돌연변이입니다. 이 효소는 exonuclease 활성이 없어 random priming protocol에 사용하기 이상적이며 Alexa Fluor™ AHA fluorescent nucleotide의 확실한 수율과 효율적인 통합을 보장합니다. BioPrime™ Plus Array CGH Genomic Labeling Systems에서 random octamer가 template DNA에 어닐링하여 exo- Klenow 효소에 프라이밍 부위를 제공합니다. polymerase가 priming site에서 연장되어 Alexa Fluor™ AHA 변형 nucleotides (직접 라벨링) 또는 amino-allyl 변형 nucleotides (간접 라벨링)이 통합됩니다. 그 후 표지된 샘플을 정제해 오염물을 제거하고 변성, 혼성화하여 microarray를 실시하거나(직접 라벨링 시스템) Alexa Fluor™ NHS ester에 결합시켜(간접 라벨링 시스템) 혼성화합니다(그림 2).

Table 1 summarizes the components of the various BioPrime™ Array CGH Labeling Systems. The BioPrime™ Core Module is found in all standard BioPrime™ and BioPrime™ Plus Systems and Modules. It contains exo-Klenow fragment (40 U/ μl), 2.5X random primer solution (octamers), control DNA (Salmon Sperm), stop buffer, and distilled water. Store at -20°C. The BioPrime™ Plus Systems and Modules contain the Alexa Fluor™ modified dNTP mixes or amine modified dNTP mixes and the Alexa Fluor™ NHS Esters. Store at -20°C. The standard BioPrime™ and BioPrime™ Plus Array CGH Genomic Labeling Systems also contain purification columns and purification buffers. Store at room temperature. All components are guaranteed for 6 months when properly stored. See Table 1 for product component summary.

Can exo-Klenow be purchased separately from your array labeling kits?

Yes, the catalog number for the exo-Klenow is AM2008. The enzyme concentration is 5 U/µL.

Can the PureLink columns provided to purify labeled cDNA probes be purchased separately? Which stand-alone columns are compatible?

The PureLink PCR purification system (Cat. Nos. K3100-01, K3100-02) can be used to purify labeled probe. Because tubes provided with stand-alone kit are not amber, be sure to protect your probe from light if you are doing direct labeling. Please contact Technical Support for the protocol.

Can the BioPrime Array CGH Genomic Labeling System be used for SNP analysis?

We do not recommend using the BioPrime Array CGH Genomic Labeling System to amplify genomic DNA for SNP analysis because the exo-Klenow polymerase has no proofreading activity.
