이 조직 세포 용해 시약은 25 mM bicine, 150 mM sodium chloride (pH 7.6)에서 고유한 세척제를 사용해 균질화를 통해 포유류 조직 샘플에서 단백질 용약화의 효율성을 극대화합니다. 이 간단한 조성은 단백질분해효소 억제제, 염, 환원제, 킬레이트제와 함께 사용할 수 있어 매우 다양한 샘플 유형과 용해 어플리케이션에서 활용도가 높습니다. T-PER Reagent으로 조제된 세포 용해물은 리포터 분석(luciferase, beta-galactosidase, chloramphenicol acetyl transferase 등), 단백질 키나아제 분석(PKA, PKC, tyrosine kinase 등), 면역분석(웨스턴 블롯(Western Blot), ELISAs, RIAs), 단백질 정제 절차에서 직접 사용할 수 있습니다.

여러 샘플에서 균질하고 효율적인 조직 단백질을 추출하는 작업은 다양한 조직 조성과 추출 방법으로 인해 어렵습니다. 심장과 신장과 같은 섬유질 조직과 콜라겐 조직은 동결-해동을 통해서나 mortar 및 pestle 방법을 사용한 단백질 추출은 세척제를 사용하지 않으면 비효율적입니다. 반대로, 비장과 간과 같은 연조직은 보다 부드러운 추출 프로토콜을 사용해야만 최적의 결과를 얻을 수 있습니다. I only want to buy one reagent for extraction of protein from both mammalian cells and tissues. Will M-PER Mammalian Protein Extraction Reagent work for protein extraction from tissue samples?

For extraction of protein from tissue samples as well as some difficult-to-lyse mammalian cells, we recommend using T-PER Tissue Protein Extraction Reagent (Cat. No. 78510) with homogenization.

What is the basic extraction protocol using T-PER Tissue Protein Extraction Reagent?

Homogenize tissue sample in 1:20 (w/v) of tissue to T-PER Reagent, then centrifuge to pellet cell or tissue debris, and collect supernatant. For example, use a ratio of 100 mg of tissue to 2 mL T-PER Reagent. Use a smaller volume of reagent if a more concentrated protein extract is required.
