Mouse embryonic fibroblasts (MEFs) are used as a feeder layer when culturing mouse and human embryonic stem cells (ESCs) and induced pluripotent stem cells (iPSCs). They are isolated from mouse (strain ICR) embryos. The cells are expanded and inactivated by γ irradiation and cryopreserved. Prior to cryopreservation, the cells are tested for viability (>80% post-thaw viability), verified for growth arrest by plating inactivated cells at low density and monitoring for foci of cell growth after 14 days in culture, and also tested for their ability to support growth of mouse embryonic stem cells (mESCs), human embryonic stem cells (hESCs), and marker expression (Oct 4).

What is the difference between Mouse (ICR) Inactivated Embryonic Fibroblasts (Cat. No. A24903) and CF1 Mouse Embryonic Fibroblasts, irradiated (Cat. Nos. A34180, A34181)? Can they be used to reprogram fibroblasts?

Mouse (ICR) Inactivated Embryonic Fibroblasts and CF1 Mouse Embryonic Fibroblasts, irradiated are inactivated feeder layer fibroblast cells that are isolated from different mouse strains. Researchers often have a preference for one or the other. Neither of these can be used to reprogram fibroblasts.

Does the culture vessel need to be coated prior to seeding Mouse (ICR) Inactivated Fibroblasts?

Yes, the cutlure vessel needs to be coated with Attachment Factor Protein (Cat. No. S006100) at 37 degrees C for 30 min or at room temperature for 2 hours. The coated vessels can be used immediately or stored at room temperature for up to 24 hours.

What is the sex of mouse embryros from which Mouse (ICR) Inactivated Fibroblasts are isolated?

We do not determine the sex of the embryos. The embryos were pooled from several litters which are a mixture of male and female.
