The RNAqueous Total RNA Isolation Kit is for phenol-free total RNA isolation from 1–75 mg of tissue or from 102–107 cells, using a guanidinium-based lysis/denaturant and glass fiber filter separation technology.

The RNAqueous kit provides:

  • Convenient syringe-based filtration
  • Compatibility with vacuum manifolds when processing multiple samples

How it works

The RNAqueous kit uses a powerful chaotropic solution for cell lysis, followed by RNA binding to glass fiber housed in a luer-lock syringe filter. Washing and elution solutions are simply pushed through the filter using a standard syringe (not included). Vacuum filtration is not required; however, the filter units are compatible with standard vacuum manifolds for processing several samples at once.

What is the minimum RNA size that is isolated using RNaqueous kits?

RNAs less than 200 nt are lost using the columns in the RNAqeous family of kits. These kits are not suitable for isolation of 5S rRNA or tRNA. The RNAqueous-Micro Total RNA Isolation Kit does, however, offer a modified protocol to allow isolation of miRNA from the sample.

I'm using your RNAqueous Total RNA Isolation Kit , and the filter is clogging. Do you have any suggestions?

To prevent filter clogging with viscous samples or samples containing a large amount of tissue, perform a clarifying centrifugatoin at 10,000-15,000 x g and remove to a fresh tube prior to adding ethanol to the lysis/binding solution.

What are the differences in the RNAqueous kits you offer?

RNaqueous kits are based on a rapid, filter-based RNA isolation system that does not require phenol, chloroform, or other toxic organic chemicals. The RNAqueous Total RNA Isolation Kit is designed for sample sizes of 10-75 mg of tissue or 10^6 to 10^7 cells, whereas the RNAqueous-Midi Total RNA Isolation Kit is designed for tissue samples of up to 0.5 g or 10^9 cells. The RNAqueous-Micro Total RNA Isolation Kit is optimized for the purification of total RNA from micro-sized samples such as 10-500,000 cultured cells, as few as 10 microdissected cells, or up to 10 mg tissue. The RNAqueous-96 Total RNA Isolation Kit utilizes a 96-well plate format for high-throughput RNA isolation from 100 to 2 x 10^6 cells or 0.1 to 1.5 mg of tissue. The RNAqueous-4PCR Total RNA Isolation Kit is designed for RNA isolation from small tissue samples (0.5-75 mg) and small- to medium-sized cultured cell samples (approximately 100-10^7 cells).
