The Total Cholesterol (TC) Colorimetric Assay Kit (single reagent, COD-PAP method) is a highly reliable and sensitive assay designed for the quantitative measurement of total cholesterol in serum, plasma, and animal tissue. Cholesterol is a type of sterol and lipid found in cell membranes. It is an essential component of cell structure and function, but high levels of TC in the blood have been linked to an increased risk of cardiovascular disease. The measurement of TC levels (which includes free cholesterol and cholesterol esters) is an important diagnostic tool in clinical and research settings.
The assay utilizes the COD-PAP method, which involves the hydrolysis of cholesterol esters in the sample to produce free cholesterol and free fatty acids. Free cholesterol is then oxidized by cholesterol oxidase to produce 4-cholestenone and hydrogen peroxide. In the presence of 4-aminoamylpyridine and phenol, hydrogen peroxide catalyzes peroxidase to form red quinone compounds. The color depth of the generated quinone is directly proportional to the cholesterol content, which can be measured spectrophotometrically at 510 nm.
