Free fatty acids are converted to acyl coenzyme A in the presence of acyl synthase. This acyl coenzyme A then reacts with acyl oxidase to produce hydrogen peroxide. In the presence of a specific enzyme and probe, hydrogen peroxide reacts to form a fluorescent substrate. The fluorescence intensity, measured at an excitation wavelength of 535 nm and an emission wavelength of 590 nm, is directly proportional to the concentration of free fatty acids in the sample.
