The FIX & PERM™ Cell Permeabilization Kit is used in flow cytometry for fixing and permeabilizing cells in suspension, allowing access of antibody to intracellular targets without sacrificing scatter characteristics.

The FIX & PERM™ Cell Permeabilization Kit is suitable for various biological samples and has been validated for use with cell lines, whole blood, and bone marrow. The FIX & PERM™ kit has been shown to accurately identify previously undetectable intracellular markers. Unlike other fixation and permeabilization reagents available on the market, the FIX & PERM™ staining protocol leaves the morphological scatter characteristics of the cells intact.



The FIX & PERM™ formula was specifically designed to reduce background staining and allow simultaneous addition of fluorochrome-labeled antibodies during the permeabilization step.



How It Works:

The FIX & PERM™ Cell Permeabilization Kit contains 2 reagents: Fixation Medium (Reagent A) and Permeabilization Medium (Reagent B). Cells are incubated for 15 minutes at room temperature with the fixation medium, washed, and then incubated with the permeabilization medium and fluorochrome-labeled antibody.



The FIX & PERM™ Cell Permeabilization Kit is available in two sizes (50 and 200 tests). Reagent A and Reagent B are also available separately in a bulk size (1000 tests).

Is the FIX & PERM Cell Permeabilization Kit (Cat. No. GAS003, GAS004) compatible with conjugates (dyes) besides FITC and PE?

Yes, this product is compatible with other conjugated antibodies, outside of FITC and PE.

The FIX & PERM Cell Permeabilization Kit protocol does not say what volume my cells should be in when they are added to the tube. What should I do?

Your cells should be in approximately 100 µL volume. One way to do this is to aliquot the correct number of cells (10E6) and then spin them down and decant the supernatant. The residual volume should be approximately 100 µL.

Can I use the FIX & PERM Cell Permeabilization Kit to access nuclear epitopes?

Yes. Generally, you need to increase the incubation time in the permeabilization buffer (Reagent B). Here are a couple of references:

Yes. The FIX & PERM Cell Permeabilization Kit is used in flow cytometry for fixing and permeabilizing cells in suspension, allowing access of antibody to intracellular targets without sacrificing scatter characteristics. It has been validated for use in cell lines, whole blood, and bone marrow. In addition fixing and permeabilizing cells, it lyses red blood cells. It is labeled as a general purpose reagent (GPR) and can be used in clinical settings, but it is also used widely in research labs.
