Human MRP2 Vesicles are insect derived purified plasma membranes with inserted MRP2 transport protein (multidrug resistance-associated protein, ABCC2). Our Human MRP2 Vesicles can be used to evaluate test compounds and drug interactions with MRP2 in in vitro assays. MRP2 is an ATP-binding cassette (ABC) transporter that carries organic anions.

Use Human MRP2 Vesicles to:

• Assess potential for transporter-mediated drug-drug interactions
• Obtain high quality results with a large signal to noise ratios

Concept Behind ABC Transporter Vesicles
ABC transporter vesicles are easy-to-use, efficient reagents for early assessment of a drug candidate's substrate and drug interaction potential. While ABC transporters typically mediate the export of substrates out of cells, transporters expressed on these inside-out vesicles import substrates into the vesicles. It is therefore possible to quantitatively evaluate transport activity for your compound by determining the amount incorporated into the vesicles.

Clear and Reliable Results
Prepared from Sf9 cells which have been engineered to over-express specific ABC transporters, these 'inside-out' vesicles provide high levels of transporter activity with low background, giving you a clear signal if your compound is a substrate or inhibitor of a specific efflux transporter.

Transporter Vesicle Details
Human MRP2 Vesicles are part of a larger group of transporter vesicles known as ABC transporter vesicles. They are prepared from purified plasma membranes isolated from an insect cell system (Sf9 cells transfected with baculovirus) expressing MRP2. MRP2 is expressed in various organs, and is thought to play a role in excretion of various compounds from the kidney and liver, based on its localization on the canalicular plasma membrane of hepatocytes and brush border membrane of tubular epithelial cells.

The quantity supplied is 2.5 mg total protein/500 µL per vial. The protein concentration is 5 mg/mL. When our recommended protocol is followed, 50 vesicular transport assays or 125 ATPase assays can be performed with one vial of ABC Transporter Vesicles product.

What are ATP-binding cassette (ABC) Transporter Vesicles?

Prepared from Sf9 cells which have been engineered to over-express specific ABC transporters, ATP-binding cassette (ABC) Transporter Vesicles are “inside-out” vesicles designed for use in vesicular transport assays. They allow the investigation of possible interactions between specific ABC transporters and test compounds. ABC Transporter Vesicles provide high levels of transporter activity with low background, giving a clear signal if the test compound is a substrate or inhibitor of a specific efflux transporter.

While ABC transporters typically mediate the efflux of substrates from cells, transporters expressed on these inside-out vesicles import substrates into the vesicles. It is therefore possible to quantitatively evaluate transport activity for the test compound by determining the amount incorporated into the vesicles.
Note: One vial of vesicle product can perform 50 vesicular transport assays.
