신속한 무독성 분리 절차
PureLink™ FFPE RNA Isolation Kit는 자일렌(xylene) 등의 독성 유기 용매에 의존하지 않고 탈파라핀화하는 간편한 가열절차(heating procedure)를 제공합니다. 이전 FFPE 분리법으로 수일이 걸리는 RNA 분리를 30 - 40분 내에 완료할 수 있습니다(그림 1). 탈파라핀화 가열절차가 RNA template 구조를 부분적으로 보존하여 cDNA 합성이 가능하고 template 품질을 향상시켜 reverse transcription이 가능합니다.

What is the average length of amplifiable cDNA from RNA isolated from FFPE tissues?

Since the quality of RNA is dependent on the quality of FFPE tissues itself, the amplifiable cDNA length is variable. Competitors' kits have been shown to max out around 400 bp, while in our experience, with our patent-pending method in the PureLink FFPE RNA Isolation Kit, most RT-PCR products are about 400 bp to 800 bp, and can even be over 1.3 kb.

What are the advantages to using the Invitrogen PureLink FFPE RNA Isolation Kit?

The PureLink FFPE RNA Isolation Kit provides a simple and rapid method for isolating total RNA from formalin-fixed paraffin-embedded (FFPE) samples with high-quality results. A fast and easy heating procedure replaces traditional labor-intensive protocols that use toxic organic solvents for deparaffinization and give disappointing results. This reduces the time required for total RNA isolation to 30-40 minutes instead of the hours or a full day required by traditional FFPE isolation methods. In addition, the RNA isolated with the PureLink FFPE RNA Isolation Kit is of higher quality and demonstrates increased amplifiable RNA size, allowing for RT-PCR products that are longer than those obtained with competitor kits and reagents. Isolated RNA is ideal for use in all gene expression studies, including microarrays, RT-PCR, and qRT-PCR.
