TA Cloning™ Kit는 하룻밤이 걸리는 ligation 단계를 사용해 PRC 반응에서 직접 Taq-amplified PCR 산물을 클로닝하도록 만들어졌습니다. 이 kit는 pCR™ vector를 사용하며 ≥90% 재조합체 함유 삽입물로 ≥80% 재조합체를 회수합니다. pCR™ vector의 일부 편리한 기능:

  • 3'-T overhang으로 Taq-amplified PCR 산물의 직접 ligation
  • 선택한 T7 (pCR™2.1) 또는 T7 및 Sp6 (pCR™II) promoter로 in vitro RNA transcription 및 sequencing 실시
  • flanking EcoR I site의 다용도 polylinker로 삽입물 절개 용이
  • kanamycin 또는 ampicillin 선택
  • 쉬운 blue/white colony 스크리닝
  • sequencing을 위한 M13 forward/reverse primer sites

TA Cloning™ kits contain linearized pCR™2.1 vector, ExpressLink™ T4 DNA ligase, 5X ExpressLink™ T4 DNA ligation buffer, dNTPs, 10X PCR buffer, sterile water, and controls. The competent cell kits contain One Shot™ chemically competent E. coli, S.O.C. medium, and a supercoiled control plasmid.

Store One Shot™ E. coli at -80°C. Store all other components at -20°C. All reagents are guaranteed stable for 6 months when properly stored.
