SYTOX AADvanced Ready Flow Reagent is a high-affinity nucleic acid stain for the detection of dead cells in flow cytometry. The dye has excitation/emission maxima of 546/647 nm.

SYTOX AADvanced Ready Flow Reagent is a ready-to-use, direct-to-sample dead cell stain that emits red fluorescence when bound to DNA. Add 2 drops form the dropper bottle directly into 1 x 106 cells, incubate, and analyze.

The SYTOX AADvanced Ready Flow Reagent dye is spectrally similar to 7-AAD, but with rapid uptake kinetics and relatively low CVs . It penetrates cells more efficiently than 7-AAD, providing better separation of live and dead cells.

I am using SYTOX AAdvanced as a dead cell stain, but all of my cells are labeling even though I am certain that they are supposed to be alive. These are adherent cells that I have trypsinized. Why am I getting false-dead signals?

SYTOX AAdvanced labels only dead cells because it is a cell impermeant dye. The dye can only enter cells that have a compromised plasma membrane. Trypsinization may cause temporary disruption of the plasma membrane, sufficient to allow staining with a cell impermeant dye. You can reduce the “false-dead” problem by either reducing the amount of trypsin and/or reduce the incubation time for trypsinization or use a gentler dissociation reagent such as TrypLE Express, TrypLESelect reagents, or Versene. After trypsinization, wash well, and if possible, allow a recovery time in normal culture media before staining with any of the SYTOX dyes.

I would like to label only 100 µL of sample of the same cell density using a Ready Flow product. The manual notes 2 drops per 1 mL (1 X 10E6 cells/mL). May I scale down for smaller volumes?

Yes, you may scale up or down as needed, but we recommend keeping the cell density the same. There are 41 µL per drop, so 2 drops is 82 µL (for the 1 mL of sample). You need to only scale down for this 100 µL volume, which would be 8.2 µL.

What is the concentration of dye/reagent in the Ready Flow products?

The amount of dye or reagent in the Ready Flow products is proprietary. If you need to consider the exact amount of dye or reagent for your experiment, each of the Ready Flow reagents is available as a standalone product.

Can I use the ReadyProbes reagents for flow cytometry?

This is not recommended. The ReadyProbes reagents were developed for imaging applications whereas the Ready Flow reagents were optimized for flow cytometry.
