Detect coliforms by fluorogenic assay procedure from water, wastewater, and food samples with Thermo Scientific™ Remel™ EC Medium with MUG (Dehydrated).
Hajna and Perry developed EC Medium to detect Escherichia coli and other coliforms1 in water, milk, shellfish and other materials2. Feng and Hartman added 4-methylumbelliferyl- β-D-glucuronide (MUG)3 to conventional coliform assays. This improved detection rates of E. coli as fluorescence could be detected within 4-24 hours.
Escherichia coli is a common gram-negative microorganism which may be present in food and water samples. The presence of E.coli may be indicative of fecal contamination, although their ingestion does not necessarily have adverse effects on health. Nevertheless, certain strains are responsible for diarrhoeal disease and can also lead to more serious forms of illness.
EC medium with MUG is recommended by the American Public Health Association and Food and Drug Administrtion for detection of E.coli in water, wastewater and foods4,5.
EC Medium with MUG contains casein and meat peptones that supply amino acids, peptides and nitrogenous compounds required for growth of bacteria. Lactose acts as a ready source of energy and its fermentation leads to the production of gas in the fermentation vial. Sodium chloride provides essential electrolytes and maintains osmotic equilibrium. Dipotassium phosphate and monopotassium phosphate are buffers which control the pH. MUG is hydrolyzed by glucuronidase and yields a fluorescent end product, 4-methylumbelliferone.
