MRS Broth (Dehydrated)

Isolate Lactobacillus species and differentiate gram-positive cocci using Thermo Scientific™ Remel™ MRS Broth (Dehydrated). In 1960, De Man, Rogosa and Sharpe modified a medium formulated earlier by Rogosa et al¹ by eliminating tomato juice and created a medium which supported the growth of slower growing lactobacilli². MRS broth is named for de Man, Rogosa and Sharpe and has been used to differentiate certain strains of gram-positive cocci based on their ability to produce gas in this medium.^3-6.

Lactic acid bacteria are Gram-positive, catalase- and oxidase-negative and are fastidious in their nutritional requirements. Growth is enhanced considerably by microaerobic conditions. Generally, the lactic acid bacteria show delayed growth and smaller colony size than other microorganisms. They may be overgrown in non-selective media, especially if incubation is required for 2-4 days.

MRS Agar is designed to encourage the growth of lactic acid bacteria, which includes species of the following genera: Lactobacillus, Streptococcus, Pediococcus, and Leuconostoc. All these species can produce lactic acid in considerable amounts.

• Can be used to differentiate certain strains of gram-positive cocci.
• Supports the growth of slower growing lactobacilli.
• Ammonium citrate and sodium acetate inhibit the growth of certain microorganisms, gram-negative bacteria, and molds.

This medium contains gelatin peptone and beef extract that supply essential nutrients and amino acids necessary for growth of lactobacilli. Yeast extract is a source of B-complex vitamins and dextrose provides an energy source. Polysorbate 80 supplies fatty acids required for the metabolism of lactobacilli. Ammonium citrate and sodium acetate inhibit the growth of certain microorganisms, gram-negative bacteria, and molds.
