La proteasa AcTEV™ reconoce específicamente una secuencia de siete aminoácidos (Glu-Asn-Leu-Tyr-Phe-Gln-Gly, con disociación entre Gln y Gly), por lo que es útil para eliminar etiquetas de afinidad de proteínas de fusión. La proteasa AcTEV™ es una versión mejorada de la proteasa del virus de grabado del tabaco (TEV) que es muy específica del centro, muy activa y significativamente más estable que la proteasa nativa del TEV, lo que resulta en una mayor actividad a largo plazo. Características de la proteasa AcTEV™:

Definición de unidad Una unidad de proteasa AcTEV™ disocia el 85 % de un sustrato de control de 3 µg en 1 h a 30 °C.
Condiciones de reacción de la unidad 50 mM de Tris-HCl (pH 8,0), 0,5 mM de EDTA, 1 mM de DTT, 3 µg de sustrato de control y 1 unidad de enzima en 30 µl durante 1 h a 30 °C. La proteasa AcTEV™ se prueba funcionalmente para comprobar la ausencia de cualquier actividad de proteasa inespecífica.

Why does AcTEV Protease require DTT?
A final concentration of 1 mM DTT is required for the AcTEV protease reaction. The DTT serves as a stabilizer of secondary structure, i.e., it ensures that the enzyme does not undergo oxidation. If you are unable to use DTT due to a column purification after digestion, you can leave it out and still see a successful AcTEV digestion, however we recommend using it.
No rigorous quantitative data for activity in the absence of DTT and EDTA has been collected in-house. Incubations using reaction buffer +/-DTT and +/- EDTA; have been performed, but the products were analyzed only at t=0 and t=2 hr at 30 degrees C. No difference in the amount of product generated +/-DTT or +/-EDTA was noticed, but it is possible that the detailed kinetics in a time course reaction may be a little different.
