El DMEM/F-12 avanzado (medio Eagle modificado de Dulbecco/F-12 de Ham) se utiliza por lo general como medio basal que permite el cultivo de células de mamíferos con suplementación de suero fetal bovino (SFB) reducido. En comparación con el DMEM/F-12 clásico, la suplementación sérica puede reducirse entre un 50 % y un 90 % sin ningún cambio en la tasa de crecimiento o la morfología. Entre las células cultivadas con éxito en el DMEM/F-12 avanzado, sin adaptación, se incluyen MRC-5, SP2, Vero, WI-38 y Jurkat.

Uso del DMEM/F-12 avanzado
El medio DMEM/F-12 avanzado es distinto a otros medios debido a adición de los siguientes ingredientes para permitir la reducción de suero: etanolamina, glutatión, ácido ascórbico, insulina, transferrina, albúmina de suero bovino rica en lípidos AlbuMAX™ II para cultivo celular, y los oligoelementos selenito sódico, metavanadato amónico, sulfato cúprico y cloruro manganoso. El DMEM/F-12 avanzado requiere la suplementación con entre un 1 % y un 5 % de suero fetal bovino y 4 mM de suplemento de L-glutamina o GlutaMAX™. Muchas líneas de células no requieren adaptarse a este medio La concentración de SFB debe optimizarse para cada línea de células para obtener la máxima reducción de suero. El medio DMEM/F-12 avanzado utiliza un sistema de tampones de bicarbonato sódico (3,7 g/l) y, por tanto, requiere un ambiente con un 5–10 % de CO2 para mantener el pH fisiológico.

Can NSCs generated using PSC Neural Induction Medium be expanded in the PSC Neural Induction Medium or do they need to be in the recommended NSC expansion medium, as stated in the product manual?

We recommend using neural expansion medium with the following composition to expand NSCs generated using the PSC Neural Induction Medium: - 50:50 Neurobasal medium: Advanced DMEM/F12 plus PSC Neural Induction Supplement.

PSC lines can behave differently, and we recommend performing your own validation experiments if you want to use PSC Neural Induction Medium, or you do not observe good expansion using the Neurobasal and Advanced DMEM/F12 combination.

We do not offer DMEM/F12 medium without folic acid as a standard catalog product. However, our Customs group can make it for you as a custom media. What is the difference between Advanced DMEM/F12 Medium and DMEM/F12 Medium?

Advanced Media are widely used basal media, which require 50-90% less Fetal Bovine Serum (FBS) supplementation compared to conventional basal media formulations. The Advanced media is unique due to the addition of the following ingredients to allow for serum reduction: ethanolamine, glutathione, ascorbic acid, insulin, transferrin, AlbuMAX I lipid-rich bovine serum albumin for cell culture, and the trace elements sodium selenite, ammonium metavanadate, cupric sulfate, and manganous chloride.

How long can I keep my media after supplementing with serum?

Generally speaking, media can be used for up to three weeks after supplementation with serum. There are no formal studies to support this, but it is the rule of thumb used by our scientists.

My medium was shipped at room temperature but it is supposed to be stored refrigerated. Is it okay?

We routinely ship media that require long-term storage in the refrigerator at room temperature. We have done studies on representative media formulations to show that media can be at room temperature for up to a week without a problem.
