The HT 3' IVT PLUS Consumables for Biomek™ FXP Target Prep Express Kit are used in conjunction with either GeneChip™ 3' IVT PLUS Reagent Kit or GeneChip™ HT 3' IVT PLUS Reagent Kit to allow for expression analysis on our 3' IVT arrays

GeneChip™ HT 3' IVT PLUS Reagent Kit (HT 3' IVT PLUS Kit) is designed for automation of the target preparation process for expression analysis on 3' IVT arrays. Built with reagent chemistries identical to GeneChip 3' IVT PLUS Reagent Kit, this high-throughput (HT) solution allows the user to generate hybridization-ready targets from input RNA with minimal hands-on time. Used in combination with the Beckman Coulter™ Biomek FXP Target Prep Express (TPE) System, HT 3' IVT PLUS Kit is capable of processing up to 96 samples in a single run.

The kit includes two amplification kit modules coupled with hybridization and controls kits for preparing hybridization-ready targets from total RNA. The kit is a complete target preparation solution that provides:
• GeneChip 3' IVT PLUS Amplification Kit (Module 1 and Module 2)
• GeneChip Hybridization Control Kit
• GeneChip Poly-A RNA Control Kit

HT 3' IVT PLUS Kit chemistry generates amplified and biotinylated complementary RNA (cRNA, also known as amplified RNA or aRNA), from poly-A RNA in a total RNA sample. HT 3' IVT PLUS Kit uses reverse transcription by priming at the poly-A tail junction of RNA to provide gene expression profiles from mRNA. RNA amplification is based upon linear amplification and employs well-standardized T7 in vitro transcription (IVT) technology.

Features and benefits
• Automated protocol enables the processing of up to 96 samples with minimal hands-on time
• Low RNA input requirement – as little as 50 ng total RNA input
• Protocol optimization with no initial primer annealing or cDNA purification step for a streamlined workflow
• Magnetic bead purification for enhanced recovery to generate sufficient target
• Built-in controls to provide effective troubleshooting

Preguntas frecuentes

Where can I download the control sequences for expression arrays?

Sequences of all our controls can be downloaded from the sequence file that is available on product pages on thermofisher.com.

For example, here is the link to the product page for Clariom D Assay, human: https://www.thermofisher.com/order/catalog/product/902923
Go to the "Software & Data Analysis" section and under "Support Files", click on "Sequence Files: Clariom D Array, human Probe Sequences, tabular format", which you can open in Excel.

Note: If the control sequence file is not available, please contact techsupport@thermofisher.com.

Are pseudogene databases included in the design of expression arrays?

Pseudogene databases were not included in the design of expression arrays.

How long can I store labeled cDNA when working with expression microarrays?

Labeled material can be stored for 2 weeks at -20 degrees C.

What is the shelf life for expression microarrays?

Typical shelf life for expression arrays are as follows:
- All ≥ 8 µm commercial expression cartridges are stable up to 24 months.
- All ≥ 8 µm commercial resequencing cartridges are stable up to 18 months.
- All ≥ 8 µm commercial expression plates are stable up to 18 months.

Please note that this excludes products such as Exon or Gene array as they are 5 µm cartridge and plates.

Is there a recommended RIN (RNA integrity number), based on Bioanalyzer, for RNA quality for IVT and WT assays?

For IVT assays, the RIN should be 6 or higher. For WT assays, there are no recommendations based on assay priming.
