Thermo Scientific Equine Genotypes Panel 1.1 is a reagent kit for horse parentage testing and identification using short tandem repeats (STRs).
The Equine Genotypes Panel 1.1 provides all the necessary reagents for amplification of the 17 microsatellite loci. The kit contains master mix and primer mix allows co-amplification of the microsatellites in a single multiplex PCR reaction. Equine control DNA is provided to verify acceptable PCR and electrophoresis conditions. Reagents and protocols of the Equine Genotypes Panel 1.1. kit have been optimized to deliver similar peak sizes (amplification yields) for each loci. Allele callings obtained with this kit represent the true alleles of an individual, instead of 'plus-A peaks' or 'split peaks' typically interpreted when using e.g. a conventional Taq DNA polymerase. The kit employs Phusion Hot Start DNA Polymerase that has a proofreading activity and eliminates the 'plus-A peaks'. One primer from each primer pair is end-labelled with a fluorescent dye. After PCR, the fragments are separated and detected in a single electrophoresis injection, using an automated capillary electrophoresis instrument.
The Equine Genotypes Panel 1.1 allows co-amplification of the 17 microsatellites in a single multiplex PCR reaction. The Panel encompasses the following loci: VHL20, HTG4, AHT4, HMS7, HTG6, AHT5, HMS6, ASB23, ASB2, HTG10, HTG7, HMS3, HMS2, ASB17, LEX3, HMS1 and CA425. These include the nine loci recommended by the Equine Genetics and Thoroughbred Parentage Testing Standardization Committee of the International Society for Animal Genetics (ISAG) and eight additional loci commonly used for horse parentage testing and identification.

Can additional markers be added to Thermo Scientific Animal Genotyping kits?
No, extra markers cannot be added to any of the Animal Genotyping kits. All primer mixes are optimized to work in a multiplex PCR. Addition of one or more primers would require optimization of the primer concentrations and the PCR protocol.
Are primers from Thermo Scientific Animal Genotyping kits available separately for specific loci?
We do not provide primers for specific locii separately. They come only in a multiplex format. If one or several loci did not amplify from the sample, faint signals can be a result of low DNA amounts or impurities. Some loci may be more sensitive towards purity of DNA used and may vary from sample to sample. On rare occasions, an animal can have a deletion in the marker region.
Do Thermo Scientific Animal Genotyping kits include an allelic ladder?
No, none of the genotyping kits contain an allelic ladder. Animal STR loci are highly polymorphic. Each allele has too many variants to make a universal allelic ladder.
