El kit InsectSelect™ Glow incluye el vector pIZT/V5-His para una expresión de alto nivel de su
gen de interés. Este vector tiene las siguientes características:

• El promotor OpIE2 de la expresión constitutiva
• El gen de resistencia al Zeocin™ para la selección rápida de líneas celulares transfectadas de forma estable
• Epítopo C-terminal V5 y secuencia de polihistidina (6xHis) para la detección con el anticuerpo anti-V5 Invitrogen y la purificación rápida con resina quelante de níquel

Además, El pIZT/V5-His expresa una fusión de la proteína fluorescente verde y la proteína de resistencia al Zeocin™
(Zeo-GFP). La proteína de fusión Zeo-GFP permite la selección rápida de líneas celulares
transfectadas con Zeocin™ y confiere un fenotipo fluorescente que simplifica la identificación de células transfectadas.

El kit InsectSelect™ Glow incluye 20 µg de cada uno de pIZT/V5-His y pIZT/V5-His/CAT superenrollados y liofilizados, 2 µg de cada uno de los primers de secuenciación directa e inversa OpIE2 liofilizados, 1 g de Zeocin™, reactivo Cellfectin™, células Sf9 y 1 l de medios para insectos GIBCO™ Grace. El kit de vectores pIZT/V5-His contiene únicamente los vectores de expresión y los primers según lo descrito anteriormente. Almacenar las células de insectos congeladas en nitrógeno líquido. Almacenar el medio y el reactivo Cellfectin™ en +4 °C. Almacenar el resto de reactivos a -20 °C. Se garantiza la estabilidad de todos los reactivos durante 6 meses si se almacenan correctamente.

Does the incubator need to be humidified when culturing insect cells?

Humidification is only necessary for monolayer cultures and suspension cultures with volumes less than 35 mL. With shaker or spinner cultures greater than 35 mL, humidification is not necessary.
