Expi293 Expression Medium is a chemically defined, serum-free, protein-free medium for growth and transfection of suspension-adapted human embryonic kidney (HEK) 293 cells. It is specifically designed as a core component to support high density culture of Expi293F cells in the Expi293 Expression System for scalable transient protein expression.

  • Supports growth of suspension 293 cultures to densities of over 1.5 x 10^7 cells/mL
  • Transfection compatible serum-free suspension culture medium enables transfection efficiencies of greater than 70% using ExpiFectamine transfection reagent
  • Enables sustained, high level expression of high density transiently transfected cultures, achieving yields of up to 1 gram per liter of recombinant protein
  • Supports growth and transfection of 293 cells in culture formats of less than 1 mL in multiwell plates to greater than 10 L in disposable bioreactors
  • Does not contain phenol red

Expi293 Expression Medium is formulated with GlutaMAX-I reagent. It is ready to use, with no supplementation required. The chemically defined formulation results in high reproducibility and lot-to-lot reliability. Expi293 Expression Medium contains no human or animal-origin components. The medium is not recommended for adherent cell cultures.

Frequently asked questions (FAQs)

Can I purify my protein from Expi293 Expression Medium using ProBond or Ni-NTA purification systems?

Expi293 Expression Medium is not directly compatible with ProBond or Ni-NTA purification systems. We recommend performing a buffer exchange or dialyzing the samples before His-tag purification.

Can I shake Expi293 cultures faster or slower than recommended?

For most flasks, there is a dropoff in expression when you go too fast or too slow because of cell shear stress or insufficient aeration. The effect is greater in plates where there is a sharper transition between static and moving fluid in the wells.

My Expi293 Expression Medium (Cat. No. A1435102) was accidentally frozen. Can I still use it?

This product should be stored at 2-8 degrees C and should not be frozen. The biggest issue with accidentally freezing this product is its solubility. If this product was accidentally frozen, we recommend placing it in a 2-8 degree environment and allowing it to slowly thaw overnight. If the product is fully thawed and shows no signs of precipitation, then it should still be usable, but we cannot guarantee effectiveness.

Does Expi293 Expression Medium contain antibiotics?

Expi293 Expression Medium does not contain antibiotics. We do not recommend the use of antibiotics, however, 5 mL/L of Antibiotic-Antimycotic (Cat. No. 15240) containing penicillin, streptomycin, and amphotericin B may be used when required, as stated on page 1 in the manual (https://assets.thermofisher.com/TFS-Assets/LSG/manuals/expi293_exp_medium_man.pdf).

Do I need to remove Anti-Clumping Agent and Pluronic F-68 from the medium while growing Expi293 cells? How about during transfection of these cells?

You can have Anti-Clumping Agent and Pluronic F-68 in the medium while growing Expi293 cells. You need to remove Anti-Clumping Agent at least 2 passages prior to transfection of Expi293 cells because it interferes with transfection. You can add it back to the medium after transfection. Pluoronic F-68 can be present during transfection as it does not interfere with transfection.
